Isaac
Isaac is the titular character of the series, reappearing from the original Binding of Isaac.

Isaac is a reference to the biblical story of The Binding of Isaac, in which Abraham is commanded by God to sacrifice his only son, Isaac, in order to prove his loyalty,
Edmund has stated that Isaac is left-handed, but in the intro Isaac is holding a pencil with his right hand.
Isaac is bald because his mother shaved his head.
Eve
Eve is the sixth unlockable character in The Binding of Isaac.
She is unlocked by making two or more deals with the devil in one playthrough.
In The Binding of Isaac: Rebirth, she is unlocked by completing 2 levels without picking up any hearts.
Eve was added in the Halloween Update on the 1st of November, 2011.
Cain
Cain is an unlockable character in The Binding of Isaac. He is unlocked by having a total of 55 coins at any one time in a playthrough (not just attaining 55 or more coins in a playthrough).
Judas
Judas is a character unlocked after beating Satan for the first time. Judas starts with only one heart container, but has The Book Of Belial upon starting as well as 3 pennies.
Lasarus
Lazarus is an unlockable character in The Binding of Isaac: Rebirth.
Blue Baby
Blue Baby, is a character unlocked by defeating Mom's Heart ten times. He starts with three soul hearts and his health meter is unique in that he cannot gain red Heart Containers.
Any that he does gain are replaced by Soul Hearts.
Samson
Samson is unlocked when the player defeats two floors in a row without taking any damage.
Samson starts with 3 hearts and Bloody Lust: Every time Samson is dealt damage, he receives a damage boost that resets when he goes down a floor.
The Lost
The Lost starts with no health and cannot increase it by any means.
Therefore, it will die from any hit.
The Lost starts with flight, Spectral tears, theD4 (if available) and Holy Mantle (if unlocked).
The Lost can take devil deals for free and activate the button in Greed Mode without taking damage.
Lilith
Lilith is a character added in The Binding of Isaac: Afterbirth expansion.
Lilith starts out blindfolded meaning she has no ability to fire tears, she instead deals damage with the incubus Familiars that follow her.
Lilith is unlocked by completing Greed Mode with Azazel.
Azazel
Azazel is a new character in The Binding of Isaac: Rebirth unlocked by making three deals with the devil in one run. Azazel starts with a short-ranged version of Brimstone, the ability to fly, the card 0 - The Fool, and three Black Heartswithout any regular heart containers; however, unlike ???, Azazel can still obtain heart containers with items.
